The human NPP family contains seven members which can be classified into two groups according to their substrate preferences. The first class comprises the nucleotide-degrading proteins NPP1 3 and 4. NPP3 (CD203c ENPP3) is expressed in multiple organs including on epithelial and mucosal surfaces and notably on basophils and mast cells. Activation of basophils by antigen-bound IgE leads to release of inflammatory mediators and rapid upregulation of NPP3 to the cell surface. This protein is in fact a common marker for diagnosing allergen sensitivity with patient basophils by flow cytometry. Basophils and mast cells mediate the response to certain pathogens as well as acute and chronic allergic reactions. Following activation these cells release ATP which further stimulates them in an autocrine manner. NPP3 upregulation serves to degrade ATP and suppress chronic allergic inflammation (but not the acute response).

The PathHunter CD86 Ligand cell line is a stable, engineered cell line designed for use with CTLA4 Checkpoint Signaling Cell Lines. When co-cultured, the CD86 expressed in this cell line will induce activity of CTLA4 receptors, which may be assayed through SH2 domain protein recruitment. This product is a companion product designed to work with Immunoglobulin Superfamily Checkpoint Assays. The cell line included overexpresses CD86 cell surface ligand. When co-cultured with the CTLA4 Checkpoint Signaling Cell Line, the CD86 ligand binds to the checkpoint receptor, inducing receptor activity. This receptor activity leads to recruitment of the relevant SH2 domain protein to the checkpoint receptor inside the receptor cell line and production of EFC signal. The PathHunter FcγRIIb clustering cell line is a stable, engineered cell line designed to increase signal produced from a Checkpoint Cell Line via antibody-mediated crosslinking of the receptors. This product is a companion product used to determine cross-linking based increases in potency of agonist antibodies. The cell line included overexpresses FcγRIIb Fcg receptor. When co-cultured with a Checkpoint Cell Line in the presence of agonist antibody, the Fcg receptor binds to the antibody, inducing receptor clustering. The induced receptor clustering leads to increased activation and downstream signaling, leading to increased EFC signal production.
